Thirty miles south of St. Marys lies Interstate 80, a major east-west corridor between New York City and Sacramento, California. State Route 255 is a well-maintained highway, which interchanges with I-80 to the south and US Route 219 to the north. US Route 120 interchanges with Route 219 about ten miles west of St. Marys. Presently US Route 219 is under consideration for upgrading to a four-lane status, with an interchange to be located five miles west of St. Marys on Route 120.

The St. Marys area is served by three local trucking companies and numerous national and regional trucking concerns. Rail freight transportation is provided by the Buffalo & Pittsburgh Railroad, Inc., which serves local industry with direct connections to both major U.S. railroads serving the east (CSXT and NS) as well as both transcontinental Canadian Railroads (CN and CP).

The Area Transportation Authority (ATA) provides local public transportation service. St. Marys is a major industrial hub for the area, and ATA provides fixed-route commuter bus services to the community from Emporium, Bennett’s Valley, Ridgway, Johnsonburg, and Wilcox in addition to local public bus service. Fullington Bus Company operates daily Trailways bus service between Buffalo and Pittsburgh.

St. Marys Airport is a modern, well-equipped facility which can accommodate most small corporate jet aircraft. The DuBois-Jefferson County Airport is located 35 miles south and the Bradford Regional Airport is 35 miles north. Both of these airports provide regularly scheduled commercial flights to major cities in the northeast.
